本文报告人低分化鼻咽癌裸小鼠移植模型的建立及其生物学特性的研究,实验动物为NC系裸小鼠(少量Swiss系),使用CNE—2 Z细胞(1×10~6/0.5ml细胞)皮下接种,历时二年,在203只动物传代30次,移植成功率为100%。组织学及超微结构显示为低分化鳞状细胞癌特征,染色体显示人类肿瘤细胞染色体,有EB病毒基因组存在及EB病毒核抗元(EBNA)、膜蛋白(LMP)表达。我们所建立人低分化鳞状细胞癌裸鼠移植模型,具有移植成功率高,呈进行性生长,恶性程度高,不会自行消退等特点,可供进一步研究鼻咽癌生物学特性及实验性治疗之用。
This article reports the establishment and biological characteristics of a poorly differentiated nasopharyngeal carcinoma transplant model in nude mice. The experimental animals were NC nude mice (a small number of Swiss lines) and used CNE-2 Z cells (1×10~6/ 0.5 ml of cells) were inoculated subcutaneously for two years, and 203 animals were passaged 30 times. The success rate of transplantation was 100%. Histology and ultrastructure showed poorly differentiated squamous cell carcinoma characteristics. Chromosome display of human tumor cell chromosomes, presence of EBV genome and expression of EB virus nuclear antigen (EBNA) and membrane protein (LMP). Our established human poorly-differentiated squamous cell carcinoma nude mice transplantation model has the characteristics of high success rate of transplantation, progressive growth, high degree of malignancy, and no self-remission, which can be used to further study the biological characteristics and experimental characteristics of nasopharyngeal carcinoma. Use for treatment.
