目的:观察肉蔻五味丸对慢性应激性抑郁症大鼠学习记忆能力及海马脑源性神经营养因子(BDNF)、酪氨酸激酶受体B(Trk B)蛋白表达的影响,探讨其抗抑郁机制。方法:将60只雄性SD大鼠随机分为正常组、模型组、阳性对照组和蒙药高、中、低剂量组,每组10只。除正常组外,对其他组大鼠进行21d的慢性应激加孤养建立模型。造模第1天起阳性对照组灌服盐酸氟西汀胶囊3.6mg/kg,蒙药高、中、低剂量组分别灌服肉蔻五味丸0.8、0.4、0.2g/kg,每日1次。应激结束时,通过Morris水迷宫实验观察大鼠学习与记忆能力改变;应用免疫组化染色法检测大鼠海马CA3区BDNF、Trk B蛋白表达情况。结果:与正常组比较,模型组大鼠Morris水迷宫逃避潜伏期延长,穿越平台次数减少,海马CA3区BDNF、Trk B蛋白表达减弱(P<0.01,P<0.05)。与模型组比较,阳性对照组和蒙药高、中剂量组大鼠Morris水迷宫逃避潜伏期不同程度缩短,海马CA3区BDNF蛋白表达增加(P<0.01,P<0.05);阳性对照组和蒙药高剂量组穿越平台次数减少,并海马CA3区Trk B蛋白表达增加(P<0.01,P<0.05)。结论:肉蔻五味丸可改善抑郁症大鼠学习记忆能力,且可能通过提高海马CA3区BDNF、Trk B蛋白表达水发挥抗抑郁作用。
Objective: To observe the effect of Rou Kou Wu Wei Wan on the learning and memory abilities and the expression of brain-derived neurotrophic factor (BDNF) and tyrosine kinase B (Trk B) in chronic stress depression rats, Depression mechanism. Methods: Sixty male Sprague-Dawley rats were randomly divided into normal group, model group, positive control group and Mongolian high, medium and low dose groups, with 10 rats in each group. In addition to the normal group, rats in other groups were subjected to chronic stress and solitary day for 21 days to establish a model. From the first day of model making, the positive control group was given fluoxetine hydrochloride capsule 3.6mg / kg, and the Mongolian medicine high, medium and low dose groups were given Rouxuweiwei pill 0.8,0.4,0.2g / kg once daily. At the end of stress, Morris water maze test was used to observe the learning and memory abilities of rats. The expressions of BDNF and Trk B protein in hippocampal CA3 area of rats were detected by immunohistochemical staining. Results: Compared with the normal group, the escape latency of Morris water maze in model group was prolonged and the number of crossing platform was decreased. The expression of BDNF and Trk B protein in hippocampal CA3 area was decreased (P <0.01, P <0.05). Compared with the model group, the escape latency of Morris water maze in the positive control group and the Mongolian high and middle dose groups was shortened to different extents, and the expression of BDNF protein in hippocampal CA3 region was increased (P <0.01, P <0.05); the positive control group and Mongolian drug high dose The number of crossing the platform decreased and the expression of Trk B protein in hippocampal CA3 region increased (P <0.01, P <0.05). Conclusion: The nutmeg wuwei pill can improve the learning and memory abilities of depression rats, and may exert the antidepressant effects by increasing the protein expression of BDNF and Trk B in hippocampal CA3 area.
