目的:探讨河南省汉族人群中基质金属蛋白酶-1(MMP-1)基因-519A/G多态性及其血浆水平与急性冠状动脉综合征(ACS)的相关性。方法:采用多聚酶链反应-限制性片断长度多态性分析(PCR-RFLP)结合琼脂糖凝胶电泳技术检测295例ACS患者、105例稳定型心绞痛(SAP)患者和198例健康对照者MMP-1基因-519A/G多态性;采用酶联免疫吸附法(ELISA)测定血浆MMP-l水平,比较2组之间MMP-1基因多态性频率分布的差异和血浆水平的差异。结果:①ACS组MMP-1基因AA型所占比例明显高于SAP组及对照组,差异具有统计学意义(P<0.05);A等位基因频率在ACS组、SAP组和对照组分别为90.5%、83.3%、83.6%,ACS组与SAP组和对照组比较,差异亦有统计学意义(P<0.05)。②ACS组血浆MMP-l水平高于SAP组和对照组(P<0.05)。③不同基因型之间血浆MMP-1水平差异无统计学意义。结论:MMP-1基因-519A/G多态性可能与河南省汉族人群ACS发病相关;血浆MMP-l水平与冠状动脉斑块不稳定性相关。
Objective: To investigate the association between the MMP-1 gene -519A / G polymorphism and its plasma level and acute coronary syndrome (ACS) in Han nationality in Henan province. Methods: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and agarose gel electrophoresis were used to detect MMP-9 in 295 ACS patients, 105 stable angina pectoris patients and 198 healthy controls. 1 gene -519A / G polymorphism. The level of plasma MMP-1 was measured by enzyme-linked immunosorbent assay (ELISA). The difference in the frequency distribution of MMP-1 gene polymorphism and the plasma level between the two groups were compared. Results: (1) The proportion of AA genotype of MMP-1 gene in ACS group was significantly higher than that in SAP group and control group (P <0.05). The frequency of A allele in ACS group, SAP group and control group were 90.5 %, 83.3% and 83.6%, respectively. There was also a significant difference between ACS group and SAP group and control group (P <0.05). ② The level of plasma MMP-1 in ACS group was higher than that in SAP group and control group (P <0.05). ③ There was no significant difference in plasma MMP-1 levels among different genotypes. CONCLUSIONS: The -519A / G polymorphism of MMP-1 gene may be associated with ACS in Henan Han population. The plasma MMP-1 level is correlated with coronary plaque instability.
