目的:观察加味苓桂术甘汤对大鼠慢性肾功能衰竭的影响。方法:将40只雄性Wistar大鼠随机分为正常对照组、模型组、低剂量组与高剂量组各10只。采用腺嘌呤构建慢性肾功能衰竭模型,低剂量组、高剂量组大鼠给予腺嘌呤后第6天实施加味苓桂术甘汤水煎液灌胃给药,给药24天后取血3mL,测量大鼠肾功能、血脂,并立即解剖取肾,10%甲醛溶液固定,通过石蜡切片、HE染色,观察肾脏眼观和镜下的病理变化。结果:生化指标结果显示,与正常对照组比较,模型组大鼠的肌酐、尿素、血脂均显著升高(P<0.01);与模型组相比,低剂量组、高剂量组大鼠的肌酐、尿素、血脂均显著降低(P<0.01),且高剂量组降低更显著(P<0.01);HE染色结果显示:与正常对照组比较,模型组大鼠的肾小管变性坏死程度较重,低剂量组、高剂量组较模型组肾小管变性坏死程度较轻。结论:加味苓桂术甘汤对延缓大鼠慢性肾功能衰竭具有一定的影响。
Objective: To observe the effects of Modified Lingguizhugan Decoction on chronic renal failure in rats. Methods: Forty male Wistar rats were randomly divided into normal control group, model group, low dose group and high dose group. Adopting adenine to construct the model of chronic renal failure, the rats of low dose group and high dose group were administrated with decoction of Lingguizhugan decoction orally on the 6th day after administration of adenine. After 24 days, Renal function, blood lipids, and immediately dissected to take the kidney, 10% formalin fixed, paraffin sections, HE staining, observe the pathological changes of kidney and microscopic observation. Results: The biochemical indexes showed that compared with the normal control group, the creatinine, urea and blood lipid in the model group were significantly increased (P <0.01). Compared with the model group, the creatinine (P <0.01), and the decrease was more significant in the high-dose group (P <0.01). HE staining showed that compared with the normal control group, the degree of renal tubular degeneration and necrosis in the model group was more severe, In the low-dose group and high-dose group, the extent of renal tubular degeneration and necrosis was lighter than the model group. Conclusion: Modified Lingguizhugan decoction has a certain effect on delaying chronic renal failure in rats.
