目的探讨补中益气颗粒对糖尿病胃肠功能紊乱大鼠的影响及其可能作用机制。方法将40只大鼠按随机数字表法分为造模组30只,正常组10只。造模组建立糖尿病胃肠功能紊乱大鼠模型,造模成功的27只大鼠随机分为模型对照组、补中益气颗粒治疗组、枸橼酸莫沙必利治疗组,每组各9只。连续灌胃7周,测定各组大鼠空腹血糖、胃残留率、血浆SP水平、胃窦SCF的蛋白质表达水平。结果与正常组比较,模型对照组大鼠空腹血糖升高、胃排空率降低、血浆SP水平减低、胃窦SCF的蛋白质表达水平降低,差异有统计学意义(P<0.05)。与模型对照组相比,补中益气颗粒组、枸橼酸莫沙必利组大鼠胃排空率均明显增加,差异有统计学意义(P<0.05);补中益气颗粒组及枸橼酸莫沙必利组大鼠血浆SP水平升高,差异有统计学意义(P<0.05);补中益气颗粒组和枸橼酸莫沙必利组大鼠SCF蛋白表达增加,差异有统计学意义(P<0.05)。结论补中益气颗粒可能通过提高大鼠血浆SP水平,提高大鼠胃窦SCF蛋白表达,增强SCF/kit信号传导,增加胃排空率,改善大鼠胃肠蠕动功能,促进胃肠动力。
Objective To investigate the effect of Buzhong Yiqi granule on diabetic rats with gastrointestinal disorders and its possible mechanism. Methods Forty rats were randomly divided into model group (n = 30) and normal group (n = 10). The model group was established diabetic rat model of gastrointestinal dysfunction, the model of successful 27 rats were randomly divided into model control group, Buzhongyiqi granule treatment group, mosapride citrate treatment group, each group 9 only. After 7 weeks of continuous gavage, fasting blood glucose, gastric residual rate, plasma SP level and SCF protein expression in gastric antrum were measured. Results Compared with normal group, fasting blood glucose, gastric emptying rate, plasma SP level and gastric SCF protein expression in model control group decreased significantly (P <0.05). Compared with the model control group, the gastric emptying rate of rats in Buzhong Yiqi granule group and mosapride citrate group were significantly increased (P <0.05), and that of Buzhong Yiqi granule group and The level of SP in the mosapride citrate group was significantly higher than that in the sham-operated group (P <0.05). The SCF protein expression was increased in the Buzhong Yiqi granule group and the mosapride citrate group, and the difference was statistically significant There was statistical significance (P <0.05). Conclusion Buzhong Yiqi Granule may improve gastrointestinal motility by increasing SP level in rat plasma, increasing SCF protein expression in gastric antrum, enhancing SCF / kit signaling, increasing gastric emptying rate, improving gastrointestinal motility in rats.
