本实验以健康孕妇尿为原料,利用苯甲酸沉淀,有机溶剂沉积,离子交换层析和凝胶过滤等方法得到HCG纯品(14000IU/mg)。用该产品免疫小鼠。将产生的免疫鼠脾脏与NS─1骨髓瘤细胞用半固相法进行杂交细胞融合,筛选纯化出HCG单抗克隆抗体(Ka=7.5×108m1)。将HCG单抗与Sepharose4B偶联制成免疫吸附亲和层析柱,可将HCG粗品(2000IU/mg)一次纯化得到HCG纯品(15000IU/mg)。建立了一种简捷、快速、经济、产品活性高的提纯HCG的新方法。
In this study, healthy pregnant women urine as raw material, the use of benzoic acid precipitation, organic solvent deposition, ion exchange chromatography and gel filtration and other methods to get HCG pure product (14000IU / mg). Mice were immunized with this product. The resulting immunized rat spleen and NS-1 myeloma cells by semi-solid phase hybridization of hybridomas, screened and purified HCG monoclonal antibody (Ka = 7.5 × 108m1). HCG monoclonal antibody coupled with Sepharose 4B affinity chromatography affinity column, crude HCG (2000IU / mg) can be purified once purified HCG product (15000IU / mg). A new method of purifying HCG was established, which was simple, rapid, economical and high in product activity.
