目的:检测乳腺癌患者外周血ALDH1highCD44+CD24-/low细胞含量的变化,探讨其与乳腺癌发生的关系及临床意义。方法:观察对象为46例Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期乳腺癌患者和12名健康女性志愿者(对照组)。采用流式细胞仪检测、分析各组外周血中ALDH1high细胞和ALDH1highCD44+CD24-/low细胞含量及其变化情况。结果:①所有样本均能检出ALDH1high细胞,各组间均数比较差异无统计学意义(P>0.05);②Ⅳ期乳腺癌组的外周血ALDH1highCD44+CD24-/low细胞含量与其他各组比较差异均有统计学意义(P<0.05),而其他各组间比较差异无统计学意义(P>0.05);③Ⅳ期乳腺癌患者原发癌灶免疫组化结果各不相同,但均可在外周血中检出ALDH1highCD44+CD24-/low细胞。结论:①各组中的ALDH1high细胞含量无显著差异,说明ALDH1具有普遍干细胞性质;②ALDH1highCD44+CD24-/low细胞含量与乳腺癌的发生有明显相关性。③ALDH1highCD44+CD24-/low细胞与乳腺癌的免疫分型无相关性。
Objective: To detect the changes of ALDH1 high CD44 + CD24- / low cells in peripheral blood of patients with breast cancer, and to explore its relationship with the occurrence of breast cancer and its clinical significance. Methods: Forty-six patients with stage I, II, III and IV breast cancer and 12 healthy female volunteers (control group) were observed. Flow cytometry was used to detect the content of ALDH1high cells and ALDH1highCD44 + CD24- / low cells in peripheral blood of each group and their changes. Results: ① ALDH1high cells were detected in all the samples, and there was no significant difference between the groups (P> 0.05); ② The content of ALDH1highCD44 + CD24- / low cells in the peripheral blood of the patients with stage Ⅳ breast cancer was significantly higher than that of other groups (P <0.05), but there was no significant difference among the other groups (P> 0.05); ③ The immunohistochemical results of primary foci in patients with stage Ⅳ breast cancer were different, ALDH1 high CD44 + CD24- / low cells were detected in peripheral blood. Conclusion: ① There is no significant difference in the content of ALDH1high cells in each group, indicating that ALDH1 has universal stem cell properties; ②HLAH1highCD44 + CD24- / low cell content has a clear correlation with the occurrence of breast cancer. ③LDH1highCD44 + CD24- / low cells and breast cancer immunophenotype no correlation.
