为了增强抗体的效应功能 ,人们尝试多种方法改造抗体分子 ,双特异性抗体是改善抗体治疗效果的发展方向之一 ,现已成为抗体工程研究领域的热点。随着分子生物学技术的迅速发展 ,出现了基因工程双特异性抗体的多种构建模式 ,并且有多种基因工程双特异性抗体制剂已经用于肿瘤的临床诊断和治疗试验。本文对基因工程双特异性抗体的构建模式按照双特异性微抗体、双链抗体、单链双特异性抗体和多价双特异性抗体 4类进行了综述。
In order to enhance the function of antibody, many attempts have been made to transform antibody molecules. Bispecific antibody is one of the development directions to improve the therapeutic effect of antibody, and has become a hot spot in the field of antibody engineering. With the rapid development of molecular biology techniques, various construction modes of genetically engineered bispecific antibodies have emerged and a number of genetically engineered bispecific antibody preparations have been used in the clinical diagnosis and treatment trials of tumors. In this paper, the construction of genetically engineered bispecific antibodies are reviewed in accordance with the four categories of bispecific, diabody, single chain bispecific and multivalent bispecific antibodies.
