利用采自山东章丘、济宁表现典型粗缩病症状的玉米植株样品进行ELISA和RT-PCR检测。间接ELISA结果表明,二者均与水稻黑条矮缩病毒(RBSDV)多克隆抗体表现阳性反应。RT-PCR扩增出该病毒基因组S10约1.8 kb片段。获得的1 801 bp全长序列与已发表的相关序列进行同源性分析和系统进化树分析,结果表明,章丘分离物克隆JNZ-14序列、济宁分离物克隆JN-8序列均属于RBSDV,但其与玉米粗缩病毒(MRDV)的进化关系比其与南方水稻黑条矮缩病毒(SRBSDV)的关系还要近,且2个分离物分别处于属于RBSDV的Ⅱ或Ⅰ两大分支中。
ELISA and RT-PCR were performed on maize plant samples collected from Zhangqiu and Jining in China. Indirect ELISA results showed that both of them reacted positively with the polyclonal antibody of rice black streaked dwarf virus (RBSDV). The 1.8 kb fragment of the S10 genome was amplified by RT-PCR. Homology analysis and phylogenetic tree analysis of the 1 801 bp full-length sequence and the published sequence showed that the sequences of JNZ-14 and JN-8 of Jining isolates belonged to RBSDV, However, its evolutionary relationship with maize rough virus (MRDV) was closer than that with SRBSDV, and the two isolates were located in the two branches of Ⅱ or Ⅰ belonging to RBSDV respectively.
