瓜子金对哮喘模型小鼠气道炎症和气道重塑的治疗作用及机制

目的瓜子金对哮喘模型小鼠气道炎症和气道重塑的治疗作用及其机制。方法将40只BALB/C雌性小鼠,随机分四个组,分别为空白对照组、支气管哮喘模型组、瓜子金治疗组(0.2g/Kg)、糖皮质激素阳性对照组(0.5mg/Kg)。在末次激发24h后取左肺下叶组织行HE、PAS染色,观察小鼠肺组织形态结构变化。取左肺上叶组织用Western blot法检测NF-kB p65蛋白的表达情况。用酶联免疫吸附法(ELISA)检测小鼠支气管肺泡灌洗液(BALF)中白细胞介素-13(IL-13)、IL-4、IL-5含量;用Diff-Quick染色法观察BALF中细胞总数和各种炎症细胞计数。结果哮喘组与空白对照组比较BALF中细胞总数、细胞分类计数、IL-13、IL-5、IL-4的含量增高,肺组织细胞核内NF-kB p65蛋白含量表达明显升高而在细胞质内NF-kB p65蛋白含量则明显降低;而瓜子金治疗组、糖皮质激素组与哮喘模型组相比BALF中细胞总数、各个细胞分类计数、IL-13、IL-5、IL-4的含量明显降低,肺组织细胞核内NF-kB p65蛋白含量表达明显降低而在细胞质内NF-kB p65蛋白含量则明显升高。哮喘组与空白对照组比较,肺组织可见多种炎症细胞浸润,支气管腔内分泌物增多,杯状细胞增殖,气道平滑肌肌层及基底膜增厚,气道上皮细胞下纤维化等气道重塑改变;而瓜子金治疗组、糖皮质激素阳性对照组与哮喘组比较上述组织病理改变均有明显好转。结论瓜子金可能通过抑制Th2类细胞因子及NF-kB p65蛋白表达,而抑制气道炎症和气道重塑的发生、发展。 Objective To study the therapeutic effect and mechanism of melon seeds on airway inflammation and airway remodeling in asthmatic mice. Methods Forty BALB / C female mice were randomly divided into four groups: blank control group, bronchial asthma model group, guaizujin treatment group (0.2g / kg), glucocorticoid positive control group (0.5mg / kg ). 24h after the last challenge, the left lower lobe tissue was taken for HE and PAS staining to observe the morphological changes of the lung tissue of the mice. Take the left upper lobe tissue by Western blot detection NF-kB p65 protein expression. The contents of IL-13, IL-4 and IL-5 in bronchoalveolar lavage fluid (BALF) of mice were detected by enzyme-linked immunosorbent assay (ELISA) The total number of cells and various inflammatory cell counts. Results Compared with the blank control group, the expression of NF-κB p65 protein in BALF increased significantly in the BALF and increased in the cytoplasm NF-kB p65 protein content was significantly reduced; while the melon seeds gold treatment group, glucocorticoid group compared with the asthma model group, the total number of cells in BALF, each cell count, IL-13, IL-5, IL- Decreased, the expression of NF-kB p65 protein in the nucleus of the lung decreased obviously while the content of NF-κB p65 protein in the cytoplasm increased obviously. Compared with the control group, asthmatic group showed a variety of inflammatory cell infiltration, increased bronchial secretions, goblet cell proliferation, airway smooth muscle and basement membrane thickening, airway epithelial fibrosis and other airway weight Plastic changes; and melon seeds gold treatment group, glucocorticoid positive control group and asthma group compared to the above histopathological changes were significantly improved. Conclusion The melon seed gold may inhibit the occurrence and development of airway inflammation and airway remodeling by inhibiting the expression of Th2 type cytokines and NF-κB p65 protein.
