目的通过观察重复经颅磁刺激(rTMS)在动物实验中的作用效果,探讨rTMS的安全性。方法选用健康成年Wistar大鼠36只,雌雄各半,体重250~300g,以随机数字表法分为3组,每组12只,分别为正常对照组(NC)、低频磁刺激组(TMS1)和高频磁刺激组(TMS2)。行重复低频经颅磁刺激和重复高频经颅磁刺激后,对接受不同的刺激频率和刺激量组经颅磁刺激大鼠的行为、组织病理形态学、血清髓鞘碱性蛋白(MBP)及神经元特异性烯醇化酶(NSE)含量进行观察。结果TMS1组和TMS2组血清MBP含量分别为(8.22±2.92),(7.31±2.02)μg/L,与NC组比较差异无显著性意义(P>0.05);TMS1组和TMS2组血清NSE含量分别为(0.68±0.09),(0.69±0.13)μg/L,与NC组比较差异无显著性意义(P>0.05)。TMS1组和TMS2组在刺激过程中均未出现异常活动,无肢体强直、阵挛等,脑组织形态学包括大体观察、普通光镜及电镜改变不明显。结论在一定强度和频率内重复经颅磁刺激是比较安全的。
Objective To observe the effect of rTMS in animal experiments by repeated transcranial magnetic stimulation (rTMS) to explore the safety of rTMS. Methods Thirty-six healthy adult Wistar rats were randomly divided into 3 groups (n = 12), with normal male and female, weighing 250-300g. They were normal control group (NC), low frequency magnetic stimulation group And high frequency magnetic stimulation group (TMS2). After repetitive low-frequency transcranial magnetic stimulation and repetitive high-frequency transcranial magnetic stimulation, we observed the behavior of transcranial magnetic stimulation rats with different stimulation frequency and stimulus volume, histopathology, serum MBP, And neuron-specific enolase (NSE) levels were observed. Results The serum levels of MBP in TMS1 group and TMS2 group were (8.22 ± 2.92) and (7.31 ± 2.02) μg / L, respectively, which were not significantly different from those in NC group (P> 0.05) (0.68 ± 0.09) and (0.69 ± 0.13) μg / L, respectively. There was no significant difference between the two groups (P> 0.05). TMS1 group and TMS2 group had no abnormal activity during stimulation, no limb rigidity, clonus and so on. The morphology of brain tissue including general observation, common light microscope and electron microscope did not change obviously. Conclusion It is safer to repeat transcranial magnetic stimulation in a certain intensity and frequency.
