目的 :研究XCT-790对沉默Dickkopf-1(Dkk1)腺病毒转染MG-63细胞活性和OPN、Lrp5、BMP2的影响。方法 :构建沉默Dkk1腺病毒载体,实验分为空白对照组、沉默Dkk1组、XCT-790处理空载腺病毒组、XCT-790处理沉默Dkk1组,根据组别不同分别用上述重组腺病毒转染MG63细胞,观察细胞增殖及OPN、Lrp5、BMP2表达情况。结果 :(1)与空白对照组比较,沉默Dkk1组和XCT-790处理空载腺病毒组MG63细胞活性增强,OPN、Lrp5、BMP2的表达量升高((P<0.05);(2)与Dkk1沉默组比较,XCT-790处理Dkk1沉默组的MG63细胞活性减弱,OPN、Lrp5、BMP2的表达量降低(P<0.05)。结论 :XCT-790可以降低因DKK1沉默而增强的MG63细胞活性,可以降低因DKK1沉默而增高的OPN、Lrp5、BMP2的表达量。
OBJECTIVE: To study the effect of XCT-790 on the activity of DML-1 (Dkk1) adenovirus transfected MG-63 cells and the effects of OPN, Lrp5 and BMP2. Methods: The silencing Dkk1 adenovirus vector was constructed and divided into blank control group, silencing Dkk1 group, XCT-790 empty vector group and XCT-790 silencing Dkk1 group. The recombinant adenovirus vector was transfected with the recombinant adenovirus MG63 cells to observe the cell proliferation and OPN, Lrp5, BMP2 expression. Results: (1) Compared with the blank control group, MG63 cells in silencing Dkk1 group and XCT-790 group increased the expression of OPN, Lrp5 and BMP2 (P <0.05); (2) Compared with Dkk1 silencing group, XCT-790 attenuated the activity of MG63 cells in Dkk1 silencing group and decreased the expression of OPN, Lrp5 and BMP2 (P <0.05) .Conclusion: XCT-790 can reduce the activity of MG63 cells enhanced by DKK1 silencing, Can reduce the increased expression of OPN, Lrp5, BMP2 due to DKK1 silencing.
