目的:优选黄芪药材可溶性蛋白质的最佳提取工艺。方法:对黄芪药材蛋白质提取条件进行单因素考察,并采用L9(34)正交试验设计法,采用考马斯亮蓝染色法,以黄芪药材可溶性蛋白质含量为指标,考察料液比、pH值、提取温度、提取时间四个因素对可溶性蛋白质提取率的影响,从而确定黄芪药材蛋白质的最佳提取工艺。结果:建立了黄芪药材蛋白质的最佳提取工艺:向黄芪药材粉末中加入20倍量pH为12的蒸馏水,在温度30℃条件下回流提取10 min。结论:优化的提取工艺稳定、合理、可行,能正确反映黄芪药材中的蛋白质含量,为黄芪生物活性物质的进一步研究提供了实验依据。
Objective: To optimize the extraction process of soluble protein from Radix Astragali. Methods: The extraction conditions of Astragalus membranaceus protein were investigated by single factor. The L9 (34) orthogonal design method was adopted. Coomassie brilliant blue staining was used to determine the content of soluble protein in Astragalus membranaceus. Temperature, extraction time of four factors on the extraction rate of soluble protein to determine the best Astragalus extract protein extraction process. Results: The optimal extraction process of the protein of Astragalus membranaceus was established: 20 times the amount of distilled water with pH 12 was added to Astragalus membranaceus powder and refluxed for 10 min at 30 ℃. Conclusion: The optimized extraction process is stable, reasonable and feasible, and can correctly reflect the protein content of Astragalus membranaceus, which provides an experimental basis for the further study of bioactive substances of Astragalus.
