日本血吸虫卵在磷酸盐缓冲盐水(PBS)中制成匀浆。离心后上清液为水溶性抗原(JSEA)。沉淀用8M尿素提取,得尿素溶解性抗原(JEU)。用动力学酶标固相免疫方法(K-ELISA)进行测定,JEU的抗原总活力得量为JSEA的2.83倍。将JEU通过Bio-Gel A 50 m柱分得的JEG_3为稳定的不含尿素的高抗原活力成分。作为诊断用抗原,应用K-ELISA对血吸虫病人血清进行测定,JEG_3较JSEA为敏感。SDS-聚丙烯酰胺凝胶电泳,凝胶区带电泳转移酶标免疫印斑技术,薄膜聚焦电泳和高压液相色谱分析的结果均表明,JSEA和JEG_3含有不同的蛋白质抗原成分,前者各组分的分子量在20万道尔顿以下,后者则主要为大分子蛋白质,分子量在40万道尔顿以上。
Schistosoma japonicum eggs are homogenized in phosphate buffered saline (PBS). The supernatant after centrifugation is water-soluble antigen (JSEA). The kinetic enzyme-linked immunosorbent assay (K-ELISA) was used to determine the total antigen yield of JEU, which was 2.83 times that of JSEA. Results of SDS-polyacrylamide gel electrophoresis, gel electrophoresis and enzyme-linked immunoblot blotting, thin-film polyacrylamide gel electrophoresis and high-pressure liquid chromatography showed that JSEA and JEG_3 contained different protein antigen components, Of the molecular weight of 200,000 Daltons or less, while the latter is mainly a macromolecular protein, the molecular weight of more than 400,000 Daltons.
