目的:建立一种可用于莽草酸及其衍生物定性检测的高灵敏度荧光薄层色谱方法。方法:含有莽草酸及其衍生物的甲醇溶液经硅胶薄层层析展开后喷洒0.1%的高锰酸钾-1%硫酸水溶液,105℃加热20min后在365nm下观测荧光。结果:莽草酸及其单、双、三取代后的衍生物在365nm下均可观测到蓝绿色荧光,检出限可达0.7mg(4.02nmol)。结论:本研究为莽草酸及其衍生物的定性鉴定提供了一种快速、灵敏、简便的方法,可广泛用于莽草酸及其衍生物的分析和检测。
OBJECTIVE: To establish a high sensitivity fluorescence thin layer chromatography method for the qualitative detection of shikimic acid and its derivatives. Methods: Methanol solution containing shikimic acid and its derivatives was developed by thin-layer chromatography on silica gel and then sprayed with 0.1% potassium permanganate-1% sulfuric acid solution. After heating at 105 ℃ for 20 min, fluorescence was observed at 365 nm. Results: The shikimic acid and its derivatives after single, double or triple substitution showed blue-green fluorescence at 365 nm with a detection limit of 0.7 mg (4.02 nmol). Conclusion: This study provides a rapid, sensitive and convenient method for the qualitative identification of shikimic acid and its derivatives. It can be widely used in the analysis and detection of shikimic acid and its derivatives.
