目的:以聚酰胺柱色谱分离,用分光光度法测定莲子心药材中的总黄酮含量。方法:以芦丁为对照品,在室温下,用NaNO2和A l(NO3)3在NaOH碱性介质中显色后,于510 nm处测定莲子心药材中总黄酮的吸光度。结果:测得6种莲子心药材中总黄酮的含量,方法在线性8.00~40.00μg/m l范围内,r=0.9992;加标回收率为98.0%~101.6%,RSD为1.8%~3.8%。结论:样品提取后用聚酰胺柱色谱分离纯化后可直接进行测定,方法简便、快速,可用于测定莲子心药材中的总黄酮含量。
OBJECTIVE: To determine the content of total flavonoids in lotus seeds by spectrophotometry with polyamide column chromatography. Methods: The absorbance of total flavonoids in lotus seeds was determined at 510 nm with rutin as a reference substance at room temperature with NaNO2 and Al (NO3) 3 in NaOH alkaline medium. Results: The content of total flavonoids in six kinds of lotus seeds was measured. The linear range was from 8.00 to 40.00 μg / ml, r = 0.9992. The recoveries were 98.0% -101.6% and RSD were 1.8% -3.8%. Conclusion: The sample can be directly separated and purified by polyamide column chromatography. The method is simple, rapid and can be used for the determination of total flavonoids in the lotus root medicine.
