目的 建立血及组织中美洛昔康药物浓度的高效液相色谱 (HPLC)检测法。方法 采用高效液相色谱法联用紫外检测器 ,检测血及组织中美洛昔康药物浓度。流动相∶甲醇∶水∶冰乙酸∶乙腈 (6 0 0∶5 0 0∶2 0∶5 0 )V/V ;检测波长 :35 5nm。结果 血及组织中美洛昔康最低检测浓度 0 0 39mg·L-1,检测限 1 95ng ,Mel在血及组织中能得到很好的分离 ,线性范围为 0 0 39~ 10mg·L-1,回归方程 :Y =3132 81 9X +4 414 3,r =0 9998,P <0 0 5 ,回收率在 85 6 7%~ 95 0 9%之间 ,日间、日内的变异均在5 6 %以下 ,足爪局部回收率在 80 13%~ 94 5 6 %,符合生物样品分析的要求。结论 本法操作简便 ,灵敏度高 ,准确性好。
Objective To establish a high performance liquid chromatography (HPLC) method for the determination of meloxicam in blood and tissues. Methods High performance liquid chromatography with UV detector was used to detect the concentration of meloxicam in blood and tissues. Mobile phase: methanol: water: glacial acetic acid: acetonitrile (60 0: 5 0 0: 2 0: 5 0) V / V; detection wavelength: 35 5nm. Results The lowest detection concentration of meloxicam in blood and tissues was 0.039 mg · L-1 with a detection limit of 1 95 ng. Mel was well separated in blood and tissues with a linear range of 0 0 39-10 mg · L -1 , The regression equation was: Y = 3132 81 9X +4 414 3, r = 0 9998, P <0 05, and the recoveries ranged from 85 6 7% to 95 0 9% %, The partial recovery of paws was 80 13% ~ 94 56%, which met the requirements of biological sample analysis. Conclusion This method is simple, sensitive and accurate.
