目的:研究肿瘤坏死因子α(TNF-α)在多囊卵巢综合征(PCOS)发病过程中的作用,并从肾虚血瘀的角度探讨补肾活血中药的干预作用。方法:以来曲唑诱导的PCOS大鼠模型为研究对象,通过TUNEL法比较模型组与正常大鼠卵巢颗粒细胞的凋亡情况,ELISA法检测血清TNF-α的浓度;并用补肾活血中药灌服干预,观察颗粒细胞凋亡及血清TNF-α浓度的变化。结果:来曲唑造模后的大鼠卵泡内颗粒细胞凋亡显著增多,血清TNF-α浓度亦相应上升。补肾活血中药干预后颗粒细胞凋亡受到明显抑制,血清TNF-α浓度亦相应降低。结论:PCOS模型大鼠血清TNF-α水平增高,卵巢颗粒细胞凋亡明显增加,可能与慢性炎症有关。补肾活血中药作用后,能够降低其颗粒细胞的凋亡并降低血清TNF-α浓度,对多囊卵巢综合征的发病有一定的干预作用。
Objective: To investigate the role of tumor necrosis factor-α (TNF-α) in the pathogenesis of polycystic ovary syndrome (PCOS) and to explore the effect of Chinese herb for invigorating the kidney and blood circulation from the point of kidney-deficiency and blood stasis. Methods: The model of PCOS induced by letrozole was used as the research object. The apoptosis of ovarian granulosa cells in model group and normal rat was compared by TUNEL method. The concentration of serum TNF-α was measured by ELISA. , Observe the changes of granulosa cell apoptosis and serum TNF-αconcentration. Results: The apoptosis of granulosa cells in rat follicles after letrozole modeling increased significantly, and the serum TNF-α concentration also increased accordingly. After the intervention of Bushen Huoxue medicine, the apoptosis of granulosa cells was obviously inhibited, and the serum TNF-α concentration also decreased accordingly. CONCLUSION: The serum level of TNF-α in PCOS rats increases and the apoptosis of ovarian granulosa cells obviously increases, which may be related to chronic inflammation. Bushenhuoxue Chinese medicine, can reduce the apoptosis of granulosa cells and lower serum TNF-α levels, the incidence of polycystic ovary syndrome have some intervention.
