目的:探讨USP22基因在氟尿嘧啶对肝癌细胞毒性作用中的影响。方法:用Western blotting检测肝癌细胞HepG2和BEL7402中USP22基因的表达状态,构建并对肝癌细胞转染USP22基因的siRNA(siRNAUSP22),然后以氟尿嘧啶处理24h后,MTT检测法观察在USP22基因沉默前后,氟尿嘧啶对肝癌细胞活性的影响。结果:USP22基因在肝癌细胞中显著高表达。siRNA-USP22转染后显著抑制USP22基因的表达,同时可明显增强氟尿嘧啶对HepG2和BEL7402细胞的毒性作用,降低其细胞活性[(53.2±6.8)%,P<0.05;(49.8±7.3)%,P<0.05]结论:下调USP22基因可提高肝癌细胞对氟尿嘧啶的敏感性,提示USP22高表达可能是造成肝癌化疗抵抗的原因之一。
Objective: To investigate the effect of USP22 gene on cytotoxicity of hepatocarcinoma with fluorouracil. Methods: The expression of USP22 gene in HepG2 and BEL7402 cells was detected by Western blotting. The siRNA targeting siRNA of USP22 gene was constructed and transfected into hepatoma cells (siRNAUSP22). After treatment with fluorouracil for 24 hours, the expression of USP22 gene was detected by MTT assay. Effect of fluorouracil on the activity of hepatocellular carcinoma cells. Results: USP22 gene was highly expressed in hepatoma cells. After transfection with siRNA-USP22, the expression of USP22 gene was significantly inhibited and the cytotoxicity of fluorouracil to HepG2 and BEL7402 cells was significantly decreased ([53.2 ± 6.8]%, P <0.05 compared with (49.8 ± 7.3)%, P <0.05] Conclusions: Down-regulation of USP22 gene can increase the sensitivity of hepatocarcinoma cells to fluorouracil, suggesting that USP22 overexpression may be one of the reasons for the chemoresistance of hepatocellular carcinoma.
