观察益气通络丹(YQTLD)含药血清对培养SD大鼠胸主动脉平滑肌细胞(VSMC)的影响 ,探讨该药临床抗冠心病、脑梗塞等心脑血管疾病的作用机制。方法血清药理学方法制备含药血清 :MTT法测定含药血清对VSMC增殖活性的影响 ;NO试剂盒测定含药血清对VSMC作用与NO的关系。结果YQTLD含药血清能显著降低培养的SD大鼠VSMCMTT法的OD值 ,与空白血清组有显著性差异(P<0.01) ;YQTLD含药血清组培养基中NO含量明显增多 ,与空白血清组有显著性差异(P<0.01)。结论YQTLD有抗VSMC增殖的作用 ,此作用可能与NO有关 ,YQTLD可能通过抗VSMC增殖活性 ,促进NO释放在临床治疗上发挥作用
Observe the effect of serum containing YQTLD on cultured rat thoracic aorta smooth muscle cells (VSMC) and explore the mechanism of the drug’s anti-cardiovascular and cerebrovascular diseases such as coronary heart disease and cerebral infarction. Methods Serum-pharmacological methods for preparation of drug-containing serum: MTT assay was used to determine the effect of drug-containing serum on the proliferation of VSMC; NO kit was used to determine the relationship between drug-containing serum and VSMC. Results YQTLD-containing serum could significantly decrease the OD value of MTT assay in cultured SD rat VSMCs, and had significant difference with the blank serum group (P<0.01). The YQTLD drug-containing serum group had significantly increased NO content in the medium, compared with the blank serum group. There was a significant difference (P<0.01). Conclusion YQTLD has anti-VSMC proliferation effect, and this effect may be related to NO. YQTLD may play a role in clinical treatment by proliferating anti-VSMC activity and promoting NO release.
