目的研究姜黄素对香烟烟雾提取物刺激巨噬细胞株RAW264.7所致炎症反应的抑制作用。方法将RAW264.7巨噬细胞株培养于含不同浓度香烟烟雾提取物或姜黄素的培养基,MTT法筛选合适的实验浓度;组蛋白去乙酰化酶抑制剂曲古抑菌素A或香烟烟雾提取物分别刺激RAW264.7细胞株,体外模拟COPD发病的炎症反应过程;应用姜黄素和/或地塞米松处理被曲古抑菌素A或香烟烟雾提取物活化的RAW264.7细胞株;ELISA法测定细胞培养上清中炎症因子TNF-α、IL-6的表达水平。结果与空白对照组比较,曲古抑菌素A或香烟烟雾提取物均可显著上调巨噬细胞株RAW264.7的TNF-α、IL-6表达;应用姜黄素或地塞米松干预,可抑制炎症因子TNF-α和IL-6的表达上调;联合应用姜黄素与地塞米松具有更强的抗炎作用。结论姜黄素可抑制香烟烟雾提取物导致的巨噬细胞炎症因子TNF-α、IL-6分泌增加,可增强地塞米松的抗炎作用,其作用机制可能与姜黄素活化组蛋白去乙酰化酶有关。
Objective To study the inhibitory effect of curcumin on inflammatory response induced by cigarette smoke extract on macrophage cell line RAW264.7. Curcumin or dexamethasone could inhibit The expression of inflammatory cytokines TNF-α and IL-6 were up-regulated. Curcumin combined with dexamethasone had a stronger anti-inflammatory effect. Conclusion Curcumin can inhibit cigarette smoke extract-induced macrophage inflammatory cytokines TNF-α, IL-6 secretion increased, can enhance the anti-inflammatory effect of dexamethasone, its mechanism of action may be related to curcumin-activated histone deacetylase related.
