研究松脂醇二葡萄糖苷(PDG)对同型半胱氨酸(Hcy)所致的血管内皮细胞ECV-304损伤的保护作用及其可能机制。Hcy损伤体外培养ECV-304建立细胞损伤模型,与不同浓度的PDG(0.1~10 g/L)共同培养36 h,检测细胞培养上清MDA、LDH、NO含量,RT-PCR法检测Caveolin-1 mRNA表达,Western blot法检测Caveolin-1的变化。与模型组相比,药物组0.1~10 g/L细胞上清MDA、LDH的含量呈浓度依赖性下降;药物组0.1~10 g/L细胞上清NO含量明显上升,并呈现一定的浓度依赖性。RT-PCR结果显示,与模型组相比,药物组0.1~10 g/L细胞的Caveolin-1 mRNA表达明显下降;用灰度扫描软件处理Western blot结果,药物组0.1~10 g/L细胞的Caveolin-1减少,与模型组比较存在显著差异。PDG拮抗Hcy诱导的血管内皮细胞ECV-304损伤,减少Caveolin-1的表达,这可能是其降血压的作用机制之一。
To study the protective effect of pinoresinol diglucoside (PDG) on vascular endothelial cell ECV-304 injury induced by homocysteine (Hcy) and its possible mechanism. Hcy injury ECV-304 cells were cultured in vitro and incubated with different concentrations of PDG (0.1 ~ 10 g / L) for 36 h. Compared with the model group, the content of MDA and LDH in the supernatant of 0.1 ~ 10 g / L drug group decreased in a concentration-dependent manner; the content of NO in the supernatant of the drug group 0.1 ~ 10 g / L increased obviously and showed certain concentration dependence Sex.
