目的探讨δ-生育三烯酚对人结肠癌SW620细胞增殖抑制作用。方法采用细胞生长曲线和real-time RT-PCR方法,观察各剂量下δ-生育三烯酚对细胞增殖抑制及对丝/苏氨酸磷酸激酶(GSK-3β)、结肠癌腺瘤性息肉蛋白(APC)及轴蛋白(Axin)在mRNA水平上表达的影响。结果δ-生育三烯酚对人结肠癌SW620细胞有明显抑制作用,24 h细胞生存率为14.4%~88.8%;GSK-3β及APC mRNA表达水平无明显变化,Axin mRNA表达水平为2.24~2.58,明显高于对照组(P<0.05)。结论δ-生育三烯酚能够明显抑制SW620细胞的生长,其机制可能与上调Axin mRNA水平有关。
Objective To investigate the inhibitory effect of δ-tocotrienol on the proliferation of human colon carcinoma SW620 cells. Methods Cell growth curve and real-time RT-PCR method were used to observe the inhibitory effect of δ-tocotrienol on the proliferation of the cells and the effect on the expression of GSK-3β, (APC) and Axin (Axin) mRNA expression levels. Results δ-Tocotrienol had a significant inhibitory effect on human colon cancer SW620 cells, with a 24 h survival rate of 14.4% -88.8%. There was no significant change in the expression of GSK-3β and APC mRNA, and Axin mRNA expression was 2.24-2.58 , Significantly higher than the control group (P <0.05). Conclusion δ-Tocotrienol can significantly inhibit the growth of SW620 cells, which may be related to the up-regulation of Axin mRNA.
