通过多相分类技术对分离自藏南地区土壤的两株根瘤菌进行了分类地位的确定。两株菌CNU8561007与CNU85000012均为革兰氏阴性、好氧的杆状细菌。通过表型特征分析发现二者在多项生理指标上表现一致,由于两菌株来源于不同的地理位置,二者在表型特征上也存在少许差异。经过比较16S rDNA序列发现,两供试菌株的序列相似性为100%,并且与Rhizobum yanglingense,R.loessense,R.mongolense,R.gallicum表现出了极高的相似性,4种看家基因atpD,recA,glnⅡ和danK的系统发育分析与16S rDNA序列分析的结果相吻合,一致显示与两供试菌株遗传距离最近的模式种为Rhizobum yanglingense,通过DNA同源性分析和DNA G+C mol%测定,最终将这两菌株归入到Rhizobum yanglingense中,两菌株在BOX指纹图谱上的差异表明了它们为同种内的不同菌株。
The taxonomic status of two rhizobia isolated from soils in southern Tibet was determined by multi-phase classification. Both strains CNU8561007 and CNU85000012 are Gram-negative, aerobic bacilli. Through the phenotypic characteristics analysis found that the two showed consistent on a number of physiological indicators, as the two strains from different geographical locations, both in the phenotypic characteristics there are also some differences. After comparison of 16S rDNA sequences, the sequence similarity of the two tested strains was 100%, and showed extremely high similarity with Rhizobum yanglingense, R. loessense, R.mongolense and R. gallicum. The four housekeeping genes atpD The phylogenetic analysis of recA, glnⅡ and danK was consistent with the results of 16S rDNA sequence analysis. The results showed that the closest genetic model to the two tested strains was Rhizobum yanglingense. DNA sequence analysis and DNA G + C mol% The two strains were eventually classified into Rhizobum yanglingense. The difference in the BOX fingerprinting of the two strains indicates that they are different isolates within the same species.
