辣根过氧化物酶(HRP)不能穿过完整的心肌肌膜,当肌膜通透性增大时则可进入肌细胞内、并能用组化呈色反应显示出来。本实验研究以HRP作示踪物,用组化方法研究喂饲克山病病区粮的大白鼠心肌细胞膜通透性的变化。 实验用雄性离乳大白鼠52只,分等量两组。一组喂饲吉林省敦化县克山病病区粮配制的饲料(病区粮组);另一组喂饲北京大兴县非病区粮配制的饲料(非病区粮组)。喂饲2个月和3个月时,每组分别取7只、16只动物,经股静脉注射HRP。心脏灌注固定,制作冰冻切片,放入Graham-Karnovsky孵育液中孵育。每组另外3只动物只注射生理盐水,作为心肌内源性过氧化物酶活性对照。用光学显微镜检查每
Horseradish peroxidase (HRP) can not penetrate the intact myocardium, and when it becomes permeable, it enters the muscle cells and can be visualized by histochemical staining. In this study, we used HRP as a tracer to study the changes of membrane permeability of rat myocardial cells fed with Keshan disease. Fifty-two male rats were randomly divided into two groups. One group was fed with feed (ward group) of Keshan disease ward in Dunhua County, Jilin Province; the other group was fed with feed (non-ward grain group) which was formulated in non-ward district of Daxing County in Beijing. Feeding two months and three months, each group were taken seven, sixteen animals, the femoral vein injection of HRP.
