目的 研究肺腺癌细胞株A5 4 9和淋巴细胞在外源性胰岛素样生长因子Ⅰ (IGF Ⅰ )作用下 ,各自IGF Ⅰ受体 (IGF ⅠR)mRNA的表达情况。方法 采用多重RT PCR对IGF ⅠRmRNA进行半定量检测 ,研究 0、5、2 0、80、16 0、32 0ng/ml不同浓度的重组人胰岛素样生长因子Ⅰ (rIGF Ⅰ )及在某一特定浓度下作用不同时间对培养的A5 4 9细胞和淋巴细胞IGF ⅠRmRNA表达的影响。结果 当IGF Ⅰ增加至 2 0ng/ml以上 ,淋巴细胞IGF ⅠRmRNA的表达显著降低 ,而A5 4 9细胞IGF ⅠRmRNA的表达未见明显改变。结论 外源性IGF Ⅰ不能显著降低肺腺癌细胞IGF ⅠR的转录水平 ,提示IGF ⅠR在肺腺癌的恶性增殖过程中可能起重要作用。
Objective To study the expression of IGF Ⅰ receptor (IGF Ⅰ R) mRNA in lung adenocarcinoma cell line A549 and lymphocytes under the action of exogenous insulin - like growth factor Ⅰ (IGF Ⅰ). Methods Multiplex RT PCR was used to detect IGF Ⅰ R mRNA semiquantitatively. The effects of different concentrations of recombinant human insulin-like growth factor Ⅰ (rIGF Ⅰ) at 0, 5, 20, 80, 160, Effects of different time on the expression of IGF Ⅰ R mRNA in cultured A549 cells and lymphocytes. Results When IGF Ⅰ increased to more than 20ng / ml, the expression of lymphocyte IGF Ⅰ RmRNA was significantly decreased, while the expression of IGF Ⅰ RmRNA in A549 cells was not significantly changed. Conclusion Exogenous IGF Ⅰ can not significantly reduce the IGF Ⅰ R transcription of lung adenocarcinoma cells, suggesting that IGF Ⅰ R may play an important role in the malignant proliferation of lung adenocarcinoma.
