探索胰岛素样生长因子(IGF)-1诱导大鼠脊髓源性神经干细胞向少突胶质细胞方向分化的机制。方法 :本研究采用Western印迹和RT-PCR方法,检测分化的神经干细胞(NSCs)中抗磷酸化细胞外信号调节激酶(ERK)1/2蛋白磷酸化的情况,以及ERK 1/2特异性抑制剂PD98059对c-fos、c-jun表达的产生的影响。结果 :胰岛素样生长因子开始表达后,NSCs中磷酸化的ERK1/2表达量逐渐增加,它的特异性抑制剂PD98059能够有效阻断这种增高作用,转录因子c-fos、c-jun的表达以及对特异性抑制剂PD98059的反应和ERK的变化保持一致。结论 :MAK-ERK信号途径在NSCs向少突胶质细胞转化的过程中起着重要作用。
