目的:研究间硝苯啶(Men)对老龄左室肥厚大鼠心肌及脑细胞膜二氢吡啶类(DHP)受体的影响.方法:选用20月龄大鼠,钳夹其左肾动脉形成肾血管高血压.采用放射配基分析法,测大鼠心肌及脑细胞膜DHP结合位点的亲和力和密度(Bmax).结果:Men20mg·kg-1·d-1ig持续9周,使老龄肾血管高血压大鼠收缩压及左室重显著降低,减低左室DHP受体总量(从595±062降到40±11pmol/LV).Men也降低丘脑和海马DHP受体Bmax(分别从522±27降到371±24pmol/gprotein和从498±26降到332±32pmol/gprotein).结论:Men在逆转老龄肾血管高血压LVH同时伴有左室,丘脑和海马钙受体数的降低.
Objective: To study the effect of m-nifedipine (Men) on the dihydropyridine (DHP) receptors in myocardium and brain cells of aged rats with left ventricular hypertrophy. Methods: 20-month-old rats were selected and their left renal artery was clamped to form renal vascular hypertension. Radioligand assay was used to measure the affinity and density (Bmax) of DHP binding sites in rat myocardium and brain cell membrane. Results: Men20mg · kg-1 · d-1ig sustained for 9 weeks, significantly decreased systolic blood pressure and left ventricular weight, and decreased the total amount of left ventricular DHP receptor in elderly hypertensive rats (from 595 ± 062 To 4 ± 1 1 pmol / LV). Men also decreased thalamus and hippocampal DHP receptor Bmax (from 522 ± 27 to 371 ± 24 pmol / gprotein and from 498 ± 26 to 332 ± 32 pmol / gprotein, respectively). Conclusion: Men can reduce the number of calcium receptors in left ventricle, thalamus and hippocampus simultaneously with LVH in elderly patients with renal hypertension.
