目的建立电纺纳米纤维为萃取介质,填装固相微萃取小柱,用于犬血浆中维拉帕米的提取净化,用高效液相色谱-紫外检测犬血浆中维拉帕米的浓度。方法采用Bea-gle犬为试验对象,口服单剂量的盐酸维拉帕米片(20mg·kg-1),0.1ml血浆样品经处理后注入固相微萃取小柱,50μl甲醇洗脱,收集洗脱液进液相色谱分析。结果维拉帕米的线性范围为50~10000μg·L-1(r=0.9999),最小检测限为25μg·L-1,提取回收率为88.1%(RSD=4.55%),精密度为4.15%~6.01%。结论此法简单方便,可替代液-液萃取法,能够用于犬血浆中维拉帕米的测定。
OBJECTIVE To establish electrospun nanofibers as extraction medium and fill solid phase microextraction cartridges for the extraction and purification of verapamil in canine plasma and determine the concentration of verapamil in the plasma of dogs using high performance liquid chromatography-ultraviolet. Methods Bea-gle dogs were used as test subjects. A single oral dose of verapamil hydrochloride (20 mg · kg -1) was given orally. 0.1 ml plasma samples were injected into solid-phase microextraction column and eluted with 50 μl methanol. Liquid off into the liquid chromatography analysis. The results showed that the linear range of verapamil was 50 ~ 10000μg · L-1 (r = 0.9999), the detection limit was 25μg · L-1, the recovery was 88.1% (RSD = 4.55%) and the precision was 4.15% ~ 6.01%. Conclusion This method is simple and convenient and can be used in the determination of verapamil in canine plasma instead of liquid-liquid extraction.
