目的:采用两种方法测定硫酸软骨素钠的相对和绝对分子质量及其分布,并对两种分析方法进行比较。方法:色谱柱为TSKgel G3000SWXL,流动相为2.84%硫酸钠(pH 5.0,含0.02%叠氮化钠),流速为0.5 mL·min-1,柱温为35℃,分子排阻色谱(SEC)法为示差折光(RI)检测器,分子排阻色谱和多角度激光光散射联用(SEC-MALLS)法的检测器为示差折光检测器(DNDC仪)和激光光散射仪联用。结果:SEC法和SEC-MALLS法测得的分子质量结果存在较大差异。结论:由于目前国内没有合适的硫酸软骨素钠分子质量测定用标样,SEC法测定结果与真实分子质量存在较大误差,SEC-MALLS法准确,可靠。
OBJECTIVE: To determine the relative and absolute molecular mass and distribution of sodium chondroitin sulfate using two methods and to compare the two methods. METHODS: The chromatographic column was TSKgel G3000SWXL with a mobile phase of 2.84% sodium sulfate (pH 5.0) containing 0.02% sodium azide and a flow rate of 0.5 mL · min-1. The column temperature was 35 ℃ and the molecular weight exclusion chromatography (SEC) The method is differential refractometer (RI) detector, molecular exclusion chromatography and multi-angle laser light scattering (SEC-MALLS) detector for the differential refractive index detector (DNDC instrument) and laser light scattering instrument. Results: The molecular mass results obtained by the SEC and SEC-MALLS methods were quite different. Conclusion: Because there is no standard sample for the determination of molecular weight of sodium chondroitin sulfate in China, there is a big error between the results of SEC and real molecular weight. The SEC-MALLS method is accurate and reliable.
