建立了血清中5-FU血药浓度的高效毛细管胶束电泳分离测定方法.测定条件为未涂渍石英毛细管(27cm*75μm);电极缓冲液(25mmol?L~(-1)硼酸-硼砂缓冲液pH9.0加50mmol?L~(-1)SDS);操作电压500V/cm;检测波长266um;压力上样10s;5-FUDR为内标.实验结果 血清中5-FU的含量在0.05~50μg?ml~(-1)之间呈良好的线性关系,r=0.999 9,平均回收率 日内回收率为98.3%(RSD=3.8%);日间为102.6%(RSR=5.2%).“,”A new method for determination of 5-FU in plasma by micellar electrokinetic capillary chromatography was established. The sample was injected with pressure of 0. 5 psi by 10s and separated in the uncoated silica capillary column (27cm*75μm) by voltage of 500v/cm: the electrophoresis buffer was 25mM boric acid-borate buffer (pH9.0) + 50mMSDS; the simple was detected by a UV-detector at 266nm with 5-FUDR as internal standard. Result: the linear range was from 0.05μg to 50μg·ml-1 (r=0.999 97) with the detection limit of 25 ng·ml-1;the average recoveries within day and between day were 98. 3% (RSD = 3.8% )and 102.6% (RSD=5.2%),respectively.
