目的:探讨玉屏风散及各组成药物体外抗炎和抗氧化损伤作用及其组成关系。方法:建立H2O2诱导红细胞膜损伤及LPS刺激人单核细胞活化的模型,应用正交试验设计表,检测不同浓度的玉屏风散及各组分对氧化损伤、炎性因子释放的抑制作用。结果:玉屏风散及各组成药物在2.5~20mg/ml的浓度范围内可明显抵抗H2O2诱导的红细胞膜损伤,抑制LPS诱导的THP-1细胞释放炎性因子TNF-α和NO,并呈良好的剂量依赖性。且应用正交试验证明了玉屏风散各组成药物在抗氧化及抗炎中的组成关系。结论:玉屏风散及各组成药物具有明显的抗炎和抗氧化活性,且黄芪起主要作用。
Objective: To investigate the anti-inflammatory and anti-oxidative injury effects of Yupingfengsan and its constituents in vitro and their relationship. Methods: The model of H2O2-induced erythrocyte membrane injury and LPS-stimulated human monocyte activation was established. The orthogonal design was used to design the model to detect the inhibitory effects of Yupufengsan and its components on the oxidative damage and the release of inflammatory cytokines. Results: Yupingfengsan and its constituents could obviously resist H2O2-induced erythrocyte membrane injury and inhibit the release of inflammatory cytokines TNF-α and NO from LPS-induced THP-1 cells in a concentration range of 2.5 ~ 20mg / ml Dose-dependent. The orthogonal experiment was used to prove the composition of Yupingfengsan in the composition of anti-oxidation and anti-inflammation. Conclusion: Yupingfengsan and its components have obvious anti-inflammatory and anti-oxidative activities, and Astragalus play a major role.
