粳稻剑叶斜下伸资源的发现与大剑叶角度的遗传和SSR标记

杂交水稻制种需对不育系割叶,此过程不但要求较高的操作技术和高强度的劳动,且对抽出的幼嫩稻穗也有伤害。本课题组在研究太湖流域粳稻地方品种资源生物多样性的过程中,发现了剑叶斜下伸的粳香糯A7444。本研究利用粳稻保持系863B(P1)与A7444(P2)进行配组,构建了P1、P2、F1、B1、B2和F2 6个世代,对剑叶角度进行分离分析。结果表明:剑叶角度受2对主基因+多基因控制,以主基因遗传为主。在分离分析的基础上,调查了P1与P2及其863B/A7444∥863B的B1世代141个单株SSR标记基因型和剑叶角度,构建该组合的SSR标记连锁图谱并分析剑叶角度QTL。利用WinQTLCart 2.5软件中的CIM方法检测到4个控制剑叶角度的QTL,其中位于第8染色体上的qFLA-8-1和qFLA-8-2为新的控制剑叶角度的主效QTL。qFLA-8-1位于SSR标记RM152-RM281之间,贡献率为55.24%,加性效应为65.33,增效等位基因来自863B,RM152在863B和A7444间扩增出的bp数分别为150 bp和145 bp,RM281在863B和A7444间扩增出的bp数分别为140 bp和145 bp。qFLA-8-2位于RM264-RM6215之间,贡献率为62.37%,加性效应为66.75,增效等位基因来自A7444,RM264在863B和A7444间扩增出的bp数分别为170 bp和167 bp,RM6215在863B和A7444间扩增出的bp数分别为185 bp和182 bp。 Hybrid rice seed production needs to cut the leaves of the sterile line. This process not only requires higher operation techniques and high-intensity labor, but also harms the young paddy that is extracted. In the process of studying the biodiversity of japonica landraces resources in the Taihu Lake Basin, the research group discovered the japonica glutinous rice A7444 with oblique extension of the flag leaf. In this study, six generations of P1, P2, F1, B1, B2 and F2 were constructed using japonica maintainer line 863B (P1) and A7444 (P2), and the angle of the flag leaf was analyzed. The results showed that the angle of flag leaf was controlled by two major genes + polygene, and the main gene was dominant. Based on the segregation analysis, SSR marker genotypes and flag leaf angles of 141 individuals of P1 and P2 and their B1 generation of 863B / A7444 ∥ 863B were investigated to construct the SSR linkage map of the combination and to analyze QTL of flag leaf angle. Four QTLs controlling the angle of flag leaf were detected by CIM method in WinQTLCart 2.5 software. Among them, qFLA-8-1 and qFLA-8-2 located on chromosome 8 were new major QTLs controlling the angle of flag leaf. qFLA-8-1 was located between SSR markers RM152-RM281 with a contribution rate of 55.24%, an additive effect of 65.33 and a synergistic allele of 863B. The number of bp amplified from RM152 between 863B and A7444 was 150 bp And 145 bp respectively. The bp numbers of RM281 amplified between 863B and A7444 were 140 bp and 145 bp, respectively. qFLA-8-2 was located between RM264 and RM6215 with a contribution rate of 62.37%, an additive effect of 66.75 and a synergistic allele of A7444. The number of bp amplified from RM264 between 863B and A7444 was 170 bp and 167 bp, the number of bp amplified from RM6215 between 863B and A7444 was 185 bp and 182 bp, respectively.
