目的:观察人类鸟苷酸结合蛋白1(GBP1)基因沉默对宫颈癌CaSki细胞增殖和迁移的影响。方法:细胞免疫荧光法检测CaSki细胞中GBP1表达。人工合成针对GBP1基因的小干扰RNA(GBP1-siRNA),体外瞬时转染CaSki细胞。Western blot法检测转染后GBP1蛋白水平变化,CCK-8法检测细胞增殖变化,Transwell小室法检测细胞迁移变化。结果:GBP1主要定位于CaSki细胞胞浆。与对照组比较,GBP1-siRNA转染后细胞中GBP1蛋白水平明显下降(P<0.01),细胞增殖能力增高(P<0.01),平均细胞穿膜数增高(P<0.001)。结论:GBP1基因沉默能促进宫颈癌CaSki细胞的增殖和迁移,GBP1基因的存在可能为宫颈癌的发生发展提供了一道防线,这将为宫颈癌的治疗提供新的思路。
OBJECTIVE: To observe the effect of human guanylate binding protein-1 (GBP1) gene silencing on the proliferation and migration of cervical cancer CaSki cells. Methods: The expression of GBP1 in CaSki cells was detected by immunofluorescence. Small interfering RNA (GBP1-siRNA) against the GBP1 gene was synthesized and transiently transfected into CaSki cells in vitro. Western blot was used to detect the change of GBP1 protein level after transfection. The cell proliferation was detected by CCK-8 assay and the cell migration was detected by Transwell chamber assay. Results: GBP1 mainly localized in the cytoplasm of CaSki cells. Compared with the control group, the GBP1-siRNA transfected cells significantly decreased the level of GBP1 protein (P <0.01), increased the cell proliferation ability (P <0.01), and increased the average number of cell penetrating membranes (P <0.001). CONCLUSION: Silencing of GBP1 gene can promote the proliferation and migration of cervical cancer CaSki cells. The presence of GBP1 gene may provide a defense against the occurrence and development of cervical cancer, which will provide a new idea for the treatment of cervical cancer.
