目的探讨被动吸烟对胎鼠海马超微结构的影响以及神经生长因子(NGF)表达的变化。方法24只成年雌性大鼠随即分为实验组与对照组,每组12只,对实验组大鼠进行被动吸烟造模。应用电镜观察胎鼠海马神经元的病理改变,采用免疫组化SP法检测海马神经元内神经生长因子表达变化。结果电镜下,被动吸烟组胎鼠神经元内线粒体空泡样变性,出现水泡样结构,核染色质边集;实验组胎鼠神经元神经生长因子平均光密度低于实验组,两组比较有显著性差异(P<0.05)。结论被动吸烟使胎鼠神经元结构破坏和神经生长因子表达下降,对胎鼠学习记忆能力可能有一定影响。
Objective To investigate the effects of passive smoking on the ultrastructure of fetal rat hippocampus and the changes of nerve growth factor (NGF) expression. Methods Twenty-four adult female rats were randomly divided into experimental group and control group, with 12 rats in each group. The rats in experimental group were given passive smoking. The pathological changes of fetal rat hippocampal neurons were observed under electron microscope. The expression of NGF in hippocampal neurons was detected by immunohistochemical SP method. Results Under electron microscope, mitochondrial vacuolar degeneration of fetal rat neurons in the passive smoking group showed blister-like structure and nuclear chromatin edge set. Mean optical density of nerve growth factor in fetal rat neurons was lower than that in the experimental group Significant difference (P <0.05). Conclusion Passive smoking reduces the structure of fetal rat neurons and the expression of NGF, which may affect the learning and memory abilities of fetal rats.
