将56只Wistar大鼠按体重随机分成4组,分别为对照组,低、高剂量甲基汞染毒组,右美沙芬(DM)预处理组。染毒28 d后处死大鼠,检测大鼠大脑皮质细胞形态学改变、Na+-K+-ATP酶活力、Ca2+-ATP酶活力、Ca2+含量、活性氧簇(ROS)含量及细胞凋亡情况。结果显示,随着甲基汞剂量的增加,病理形态损伤明显加剧;Na+-K+-ATP酶和Ca2+-ATP酶活力均明显降低;Ca2+和ROS含量及细胞凋亡率明显升高。与高剂量组比较,DM预处理组大鼠大脑皮质中上述指标均得到显著改善。提示右美沙芬对甲基汞致大鼠神经毒性有一定保护作用。
Fifty - six Wistar rats were randomly divided into four groups according to body weight: control group, low and high dose methylmercury exposure group and dextromethorphan (DM) pretreatment group. The rats were sacrificed 28 days after the operation and the morphological changes, Na + -K + -ATPase activity, Ca2 + -ATPase activity, Ca2 + content, reactive oxygen species (ROS) content and apoptosis in cerebral cortex were detected. The results showed that with the increase of methylmercury dose, the damage of pathological morphology was obviously aggravated; the activities of Na + -K + -ATPase and Ca2 + -ATPase were significantly decreased; the content of Ca2 + and ROS and the apoptosis rate were significantly increased. Compared with the high-dose group, the above indicators in the cerebral cortex of DM pretreatment group were significantly improved. Tip dextromethorphan methylmercury-induced neurotoxicity in rats has a protective effect.
