【目的】观察开心胶囊对犬心跳骤停复苏后心肌损伤的作用。【方法】健康杂种犬15条,随机分为假手术组、模型组、开心胶囊组;复制心脏骤停及复苏模型,分别在恢复自主循环后5min、30min、90min、180min由颈内静脉插管采取中心静脉血,观测各时段肌钙蛋白T(cTnT)的含量变化;心肌组织切片在光镜和电镜下观察心肌损伤的结构变化。【结果】模型组犬血清cTnT在自主循环恢复后立即升高,各时段cTnT浓度与假手术组比较均有显著性差异(P<0.05);开心胶囊组在复苏后90min和180min时段cTnT浓度值均低于模型组(P<0.05)。光镜下模型组心肌染色阳性点总面积增大,与假手术组比较有显著性差异(P<0.05);开心胶囊组阳性总面积降低,与模型组比较有显著性差异(P<0.05);电镜下开心胶囊组虽然出现细胞结构损伤,但损伤程度较模型组轻。【结论】开心胶囊对犬心跳骤停复苏后心肌损伤具有良好的防治作用。
【Objective】 To observe the effect of Kaixin capsule on myocardial injury after cardiac arrest in dogs. 【Methods】 Fifteen healthy mongrel dogs were randomly divided into sham operation group, model group, and Kaixin capsule group; cardiac arrest and resuscitation models were replicated, and the internal jugular vein was cannulated at 5 min, 30 min, 90 min, and 180 min after recovery of spontaneous circulation. Central venous blood was taken to observe the changes of troponin T (cTnT) content at each time period; myocardial tissue sections were observed under light microscope and electron microscope to observe the structural changes of myocardial injury. 【Results】 The serum cTnT level in the model group increased immediately after the recovery of spontaneous circulation. The cTnT concentration in each period was significantly different from that in the sham operation group (P<0.05). The cTnT concentration in the Kaixin capsule group was 90 min and 180 min after resuscitation. Both were lower than the model group (P<0.05). The total area of myocardial positive spots in the model group increased with light microscopy, and there was a significant difference compared with the sham group (P<0.05). The total positive area of the Kaixin capsule group decreased and there was a significant difference compared with the model group (P<0.05). Under the electron microscope, although the cell structure of the happy capsule group was damaged, the degree of damage was lighter than that of the model group. 【Conclusion】 Kaixin capsule has good preventive and therapeutic effects on cardiac injury after resuscitation in dogs with cardiac arrest.
