目的:研究siRNA对胃癌细胞BGC-823生长抑素(SOM)分泌的抑制效应。方法:设计4条针对SOM基因不同位点的寡核苷酸序列,应用RiboMAXT7体外转录合成siRNA并转染胃癌细胞系BGC-823,经RT- PCR、免疫细胞化学法检测SOM mRNA和蛋白的表达水平,筛选抑制效果最佳的序列,MTT法检测BGC-823细胞的增殖变化。结果:转染后24、48及72h,SOM基因的表达被抑制,抑制效率有差异,并呈浓度及时间依赖性。结论:体外合成siRNA抑制胃癌细胞系SOM基因的表达,增强了细胞的增殖能力。
AIM: To investigate the inhibitory effect of siRNA on the secretion of somatostatin (SOM) from gastric cancer cell BGC-823. Methods: Four oligonucleotides targeting SOM gene were designed and synthesized. SiRNA was transfected into human gastric cancer cell line BGC-823 by RiboMAXT7 in vitro. The expression of SOM mRNA and protein was detected by RT-PCR and immunocytochemistry Level, screening the best inhibition of the sequence, MTT assay BGC-823 cell proliferation. Results: At 24, 48 and 72 hours after transfection, the expression of SOM gene was inhibited and the inhibitory efficiency was different, in a concentration-dependent and time-dependent manner. Conclusion: In vitro synthesis of siRNA inhibits the expression of SOM gene in gastric cancer cell lines and enhances the proliferation of cells.
