目的:观察成年ROSA26小鼠中枢神经系统半乳糖苷酶(X-Gal)的表达与分布。方法:应用X-Gal组织化学结合尼氏染色及免疫组织化学双标技术研究β-gal在该嵌合体脑内的表达与分布特征。结果:X-Gal组织化学染色显示在该线系小鼠脑和脊髓中均广泛表达转基因β-gal活性产物,但以灰质为主。X-Gal和尼氏双重染色表明,β-gal在神经元、室管膜细胞和脉络丛上皮细胞中均有表达。免疫组化进一步显示β-gal广泛表达于神经元中,但在星形胶质细胞中则未见表达。结论:ROSA26小鼠中枢神经细胞稳定而广泛的表达β-gal,此转基因模式动物将可用于神经细胞培养、移植及其它相关神经生物学研究。
Objective: To observe the expression and distribution of galactosidase (X-Gal) in the central nervous system of adult ROSA26 mice. Methods: The expression and distribution of β-gal in the chimera brain were studied by X-Gal histochemistry combined with Nissl staining and immunohistochemistry double labeling. RESULTS: X-Gal histochemical staining showed extensive expression of the transgenic beta-gal product of activity in brain and spinal cord of this lineage mouse, but predominantly gray matter. X-Gal and Nissl double staining showed that β-gal is expressed in neurons, ependymal cells and choroid plexus epithelial cells. Immunohistochemistry further showed that β-gal is widely expressed in neurons but not in astrocytes. CONCLUSIONS: ROSA26 mouse CNS cells express stable and extensively expressed β-gal, and the transgenic animal will be useful for nerve cell culture, transplantation and other related neurobiology studies.
