目的:通过体外细胞培养技术考察落叶松脂醇(laricircsinol,以下称LA)对肿瘤细胞的增殖抑制及诱导凋亡作用。方法:采用MTT比色法、生长曲线法考察LA对肿瘤细胞增殖抑制率,荧光显微镜观察LA作用PC-3细胞后的细胞形态,流式细胞术检测LA对PC-3、KB、NB4、B16肿瘤细胞的凋亡率。结果:LA对4种肿瘤细胞均有较强的细胞毒作用,IC50分别为(0.32±0.04)、(3.92±0.09)、(0.59±0.05)、(2.17±0.11)μg.mL-1;生长曲线法结果表明LA对PC-3细胞的抑制作用呈显的著量-效关系,且在一定剂量范围内呈显著的时-效关系;荧光显微镜观察结果表明给药组细胞的细胞核改变和凋亡小体的形成;流式细胞术检测结果表明随LA浓度的增加细胞凋亡率增加,呈显著的量-效关系。结论:LA对多种肿瘤细胞均有抑制作用,诱导细胞凋亡是其作用机制之一。
OBJECTIVE: To investigate the inhibitory effect of laricin cinol (LA) on the proliferation of tumor cells and the induction of apoptosis by in vitro cell culture technique. Methods: MTT colorimetric assay and growth curve were used to investigate the inhibitory effect of LA on the proliferation of PC-3 cells. The morphology of PC-3 cells was observed by fluorescence microscopy. The effect of LA on PC-3, KB, NB4 and B16 Tumor cell apoptosis rate. RESULTS: LA had a strong cytotoxic effect on all four tumor cells with IC50 of (0.32 ± 0.04), (3.92 ± 0.09), (0.59 ± 0.05), (2.17 ± 0.11) μg.mL-1, respectively The results of curve analysis showed that LA had a significant dose-effect relationship with PC-3 cells and showed a significant time-effect relationship within a certain dose range. The results of fluorescence microscopy showed that the cell nucleus changes and apoptosis The results of flow cytometry showed that the apoptotic rate increased with the increase of LA concentration, showing a significant dose-response relationship. Conclusion: LA has inhibitory effect on many kinds of tumor cells, and inducing apoptosis is one of its mechanisms.
