目的观察急性白血病患儿骨髓单个核细胞(BMMNC)中转铁蛋白受体(TfR)、铁蛋白(FN)及自噬相关基因Beclin1、LC3表达情况,并探讨其意义。方法 46例急性白血病患儿,其中急性淋巴细胞白血病(ALL)患儿30例(ALL组)、急性髓细胞白血病(AML)患儿16例(AML组),另取同时期健康儿童16例(对照组)。取各组骨髓液制备BMMNC,采用逆转录PCR技术检测BMMNC中的TfR、FN、Beclin1、LC3 mRNA,采用Pearson相关分析分析TfR、FN分别与Beclin1、LC3的关系。结果 ALL组BMMNC中TfR、FN、Beclin1、LC3 mRNA的相对表达量较对照组高(P均<0.01)。AML组BMMNC中Beclin1、LC3 mRNA的相对表达量较对照组低(P均<0.01),TfR、FN mRNA的相对表达量较对照组高(P均<0.01)。在ALL组、AML组及对照组,TfR与Beclin1、LC3表达均呈负相关(P均<0.05),FN与Beclin1、LC3表达均呈正相关(P均<0.05)。结论 TfR、FN在急性白血病患儿BMMNC中表达升高;Beclin1、LC3在ALL中表达增高,而在AML中表达下降;随着自噬活性的增强,TfR表达下调、FN表达上调。
Objective To investigate the expression of transferrin receptor (TfR), ferritin (FN) and autophagy-related genes Beclin1 and LC3 in bone marrow mononuclear cells (BMMNC) of children with acute leukemia and to explore its significance. Methods A total of 46 children with acute leukemia were enrolled in this study. Among them, 30 (ALL) children with acute lymphoblastic leukemia (ALL), 16 children with acute myeloid leukemia (AML) and 16 healthy children (AML) Control group). BMMNCs were prepared from bone marrow of each group. TfR, FN, Beclin1 and LC3 mRNA in BMMNC were detected by RT-PCR. Pearson correlation analysis was used to analyze the relationship between TfR, FN and Beclin1, LC3 respectively. Results The relative expression of TfR, FN, Beclin1 and LC3 mRNA in BMMNC in ALL group was higher than that in control group (all P <0.01). The relative expression of Beclin1 and LC3 mRNA in BMMNC of AML group was lower than that in control group (all P <0.01). The relative expression of TfR and FN mRNA in AML group was higher than that in control group (all P <0.01). There was a negative correlation between TfR and Beclin1, LC3 expression in ALL group, AML group and control group (all P <0.05). FN was positively correlated with Beclin1 and LC3 (all P <0.05). Conclusions The expression of TfR and FN is increased in BMMNC of children with acute leukemia. The expression of Beclin1 and LC3 is increased in ALL and decreased in AML. The expression of TfR and FN is up-regulated with the increase of autophagy activity.
