目的观察益气熄风化痰法(痫宁片)对氯化锂联合匹罗卡品所致的慢性癫痫大鼠大脑海马区谷氨酸脱羧酶(GAD65)的免疫反应表达的影响,探讨其抗癫痫的可能机制。方法将70只SD大鼠随机分为空白组、模型组、阳性药对照组和痫宁片组,采用氯化锂和匹罗卡品复制慢性癫痫大鼠模型,分别给予生理盐水、丙戊酸钠、痫宁片灌胃,4周后采用免疫组化SABC法检测大鼠大脑GAD65的表达。结果阳性药对照组与痫宁片组大鼠大脑GAD65表达较模型组增强(P<0.05),痫宁片组与阳性药对照组比较,差异无统计学意义(P>0.05)。结论益气熄风化痰法能增强癫痫大鼠大脑海马区GAD65的表达,这是其抗癫痫的主要机制之一。
Objective To observe the effect of Yiqi Xifengsutan (Piannian Tablet) on the expression of glutamate decarboxylase (GAD65) in the hippocampus of rats with chronic epilepsy induced by lithium chloride and pilocarpine, Possible mechanism of epilepsy. Methods Seventy SD rats were randomly divided into blank group, model group, positive control group and Xianingning tablet group. Chronic epilepsy rat models were duplicated with lithium chloride and pilocarpine, and were given normal saline, valproic acid Sodium, Xianning tablets gavage, 4 weeks after immunohistochemical SABC method to detect rat brain GAD65 expression. Results The expression of GAD65 in the brain of rats in positive control group and Xianingning Tablet group was significantly higher than that in model group (P <0.05). There was no significant difference between Xianning Tablet group and positive control group (P> 0.05). Conclusion Yiqi Xifengsutan method can enhance the expression of GAD65 in the hippocampus of epileptic rats, which is one of the main mechanisms of its anti-epilepsy.
