目的探讨中药血尿停颗粒治疗多种以系膜增生为主要病理改变的肾小球疾病的作用机理。方法利用小鼠肾小球系膜细胞(GMC)体外培养技术,采用血清药理学、药物与肝S9共育2种加药方式,观察细胞增殖及白细胞介素-6(IL-6)、内皮素(ET)和乳酸脱氢酶(LDH)含量的变化。结果2种加药方式均显示,血尿停颗粒具有抑制系膜细胞(MC)的增殖及IL-6、ET的产生的作用,细胞毒作用不明显。结论抑制MC增殖及抑制MC产生IL-6和ET可能是血尿停颗粒治疗多种以系膜增生为主要病理改变的肾小球疾病的作用机制之一。
Objective To investigate the mechanism of traditional Chinese medicine Xueyu-stop Granules in treating various glomerular diseases with mesangial hyperplasia as the main pathological changes. Methods The mouse glomerular mesangial cells (GMC) were cultured in vitro. Serum pharmacology, drug and liver S9 co-incubation were used to observe cell proliferation, interleukin-6 (IL-6), and endothelium. Changes in the levels of ET and LDH. RESULTS: Both dosing methods showed that Heyushu Granules inhibited the proliferation of mesangial cells (MC) and the production of IL-6 and ET. The cytotoxic effect was not obvious. Conclusions The inhibition of MC proliferation and the inhibition of MC production of IL-6 and ET may be one of the mechanisms of the effect of Xueyaoting Granules in treating various glomerular diseases with mesangial proliferation as the main pathological change.
