目的:观察柴贝止痫汤、卡马西平、柴贝止痫汤与卡马西平联用3种不同干预对难治性癫痫大鼠模型行为学及P-糖蛋白(P-gp)和MDR1表达的影响。方法:采用侧脑室注射海人酸法制作癫痫持续状态后慢性难治性癫痫大鼠模型,分为模型组、西药组、中药组、中西药组、正常组、假手术组6组。给药时间3个月。以Western Blot及实时荧光定量PCR法检测治疗1、3个月海马P-gp及MDR1a/1b mRNA水平。结果:海马中P-gp、MDR1a/1b mRNA含量,模型组高于假手术组,在治疗1个月差异具有统计学意义(P<0.01,P<0.05);西药组高于模型组,差异不具统计学意义;中药组、中西药组低于模型组,其中P-gp含量在治疗1月时差异具有统计学意义(P<0.05);中西药组低于西药组,差异具有统计学意义(P<0.05)。结论:柴贝止痫汤能够抑制难治性癫痫P-gp表达,这可能通过对MDR1a/1b mRNA转录水平的影响实现。
OBJECTIVE: To observe the effects of Chaibai Zhixie Tang, carbamazepine, Chai Baizhi Huoxian Decoction and carbamazepine on the behavior and the expressions of P-gp and MDR1 in refractory epilepsy rats with three different interventions. Methods: Chronic intractable epilepsy rats were induced by intracerebroventricular injection of kainic acid into six groups: model group, western medicine group, traditional Chinese medicine group, western medicine group, normal group and sham operation group. Results: The contents of P-gp and MDR1a / 1b mRNA in hippocampus in model group were significantly higher than those in sham operation group (P <0.01, P <0.05), and the difference was statistically significant Not statistically significant; Chinese medicine group, the Western medicine group was lower than the model group, P-gp content in the treatment of 1 month difference was statistically significant (P <0.05); Western medicine group was lower than western medicine group, the difference was statistically significant (P <0.05). Conclusion: Chaibai Zhixian Decoction can inhibit the expression of P-gp in refractory epilepsy, which may be caused by the effect of MDR1a / 1b mRNA transcription.
