目的探讨S100A4基因mRNA表达与肾癌细胞分化、转移的关系。方法提取31例肾癌及正常组织配对标本的总RNA,用RT-PCR方法测定S100A4基因表达,并结合临床资料,对S100A4基因差异表达与肾癌病人临床表现的相关性进行分析研究。结果31例配对标本采用RT-PCR方法分别进行S100A4mRNA荧光检测比较,29例标本肿瘤组织中S100A4基因mRNA表达明显高于癌旁正常肾组织。其中低分化肿瘤组病人的S100A4指数高于高分化组(7.94vs5.06,P<0.001);发生转移(包括淋巴结转移和远处转移)者高于无转移者(9.61vs5.53,P<0.001);低年龄组病人(≤50岁)S100A4指数与高年龄组无明显差异(6.31vs6.66,P>0.05);肾颗粒细胞癌组与透明细胞癌组无明显差异(6.98vs6.02,P>0.05);肿瘤直径≤5cm组与>5cm组无明显差异(5.95vs6.93,P>0.05)。结论肾癌组织中S100A4基因mRNA可望作为肾细胞癌的病情发展及指导临床治疗的标记物之一;S100A4基因表达与肾细胞癌分化有关,癌组织S100A4mRNA测定有助于判定肿瘤的预后;S100A4基因差异表达与肾细胞癌转移明显相关。
Objective To investigate the relationship between the expression of S100A4 gene and the differentiation and metastasis of renal cell carcinoma. Methods The total RNA was extracted from matched samples from 31 cases of renal cell carcinoma and normal tissues. The expression of S100A4 gene was detected by RT-PCR. The correlation between S100A4 gene expression and clinical manifestations of renal cell carcinoma was analyzed. Results The expression of S100A4 mRNA in 31 cases of matched samples was compared with that of S100A4 mRNA by RT-PCR respectively. The mRNA expression of S100A4 in 29 cases was significantly higher than that in adjacent normal renal tissues. Among them, the S100A4 index in patients with poorly differentiated tumors was higher than that in patients with well-differentiated tumors (7.94 vs 5.06, P <0.001), but those with metastasis (including lymph node metastasis and distant metastasis) were significantly higher than those without metastasis (9.61 vs 5.53, P < 0.001). The S100A4 index in low-age group (≤50 years old) had no significant difference (6.31 vs6.66, P> 0.05) between S100A4 index and high-grade group; there was no significant difference between renal cell carcinoma group and clear cell carcinoma group (6.98vs6.02 , P> 0.05). There was no significant difference in tumor diameter ≤5cm and> 5cm (5.95 vs6.93, P> 0.05). Conclusion The mRNA of S100A4 in renal cell carcinoma is expected to be one of the markers for the progression of renal cell carcinoma and clinical therapy. The expression of S100A4 gene is correlated with the differentiation of renal cell carcinoma, and the determination of S100A4 mRNA in carcinoma can be helpful to determine the prognosis of the tumor. S100A4 Gene differential expression was significantly associated with renal cell carcinoma metastasis.
