目的 通过研究缺氧对胎盘绒毛分泌白介素 -1β(IL- 1β)的影响及重组IL- 1β体外对内皮细胞的损伤,探讨IL - 1β在妊娠期高血压疾病发病中的作用。方法 8例正常足月胎盘绒毛分别在常氧(21%O2)及低氧(2%O2)的条件下培养, 采用ELISA方法检测胎盘绒毛培养液中IL- 1β的含量;体外培养人脐静脉内皮细胞(HUVEC),在培养液中加入重组IL- 1β 培养5h后,流式细胞仪检测内皮细胞血管细胞粘附分子-1(VCAM 1)的表达。结果 胎盘绒毛在缺氧条件下培养分泌 IL- 1β的量显著高于常氧条件;重组IL- 1β体外可诱导内皮细胞VCAM 1的表达。结论 缺氧可能是影响胎盘绒毛产生IL -1β的一个重要因素,IL- 1β能引起血管内皮细胞的损伤,IL- 1β可能参与了妊娠期高血压疾病的病理发生过程。
OBJECTIVE: To investigate the effect of hypoxia on interleukin-1β (IL-1β) secreted by placenta and injury of endothelial cells induced by recombinant IL-1β in vitro to explore the role of IL-1β in the pathogenesis of hypertensive disorder complicating pregnancy. Methods Totally 8 cases of normal term placental villi were cultured under normoxia (21% O2) and hypoxia (2% O2) respectively. The contents of IL-1β in placental villi culture medium were detected by ELISA. Cultured human umbilical vein Endothelial cells (HUVECs) were cultured in culture medium containing recombinant IL-1β for 5 hours. Flow cytometry was used to detect the expression of vascular cell adhesion molecule-1 (VCAM 1) in endothelial cells. Results The amount of IL-1β secreted by placental villi in hypoxic condition was significantly higher than that in normoxic condition. Recombinant IL-1β induced the expression of VCAM-1 in endothelial cells in vitro. Conclusion Hypoxia may be an important factor that affects the production of IL-1β in placenta villi. IL-1β can induce the injury of vascular endothelial cells. IL-1β may be involved in the pathogenesis of hypertensive disorder complicating pregnancy.
