目的研究脾虚模型大鼠脑内干扰素γ(IFN-γ)、白细胞介素4(IL-4)活性表达,以及扶正益气中药的干预作用。方法 40只大鼠随机分成4组,正常组、模型组、治疗1组(四君子汤组)、治疗2组(玉屏风散),每组各10只。采用苦降泻下、饮食失节加劳倦过度法建立大鼠脾虚模型,采用称重法检测免疫器官指数,采用免疫组化法与原位杂交法检测下丘脑腹侧核、海马CA1区的IFN-γ和IL-4表达变化与四君子汤玉屏风散的治疗作用。结果模型组胸腺及脾脏指数明显下降,IFN-γ在海马CA1区和下丘脑腹侧核表达明显降低,IL-4在海马CA1区和下丘脑腹侧核表达明显增加;治疗组胸腺及脾脏指数明显增加,IFN-γ在海马CA1区和下丘脑腹侧核表达明显增加,IL-4在上述脑区表达明显降低。结论益气扶正中药四君子汤、玉屏风散可能通过影响免疫器官、调控细胞因子IFN-γ和IL-4活性表达而调节机体免疫功能。
Objective To study the expression of interferon γ (IFN-γ) and interleukin-4 (IL-4) in the brain of rats with spleen-deficiency syndrome and the intervention of Fuzhengyiqi. Methods Forty rats were randomly divided into 4 groups: normal group, model group, treatment group 1 (Sijunzi Decoction group) and treatment group 2 (Jupingfengsan), with 10 rats in each group. The rat model of spleen deficiency was established by using the method of bitter declining diarrhea, eating disorders of the abdomen and overworked rats. Immunohistochemistry and in situ hybridization were used to detect the index of immune organ in the hypothalamic ventral nucleus and hippocampal CA1 area, Changes of γ and IL-4 expression and the therapeutic effect of Si Jun Zi Tang Yu Ping Feng San. Results The indexes of thymus and spleen of model group were significantly decreased. The expression of IFN-γ in hippocampal CA1 area and hypothalamic ventral nucleus was significantly decreased. The expression of IL-4 in hippocampal CA1 area and hypothalamic ventral nucleus was significantly increased. The thymus and spleen index Significantly increased, IFN-γ in the hippocampal CA1 and hypothalamic ventral nuclei significantly increased expression of IL-4 in the brain area was significantly reduced. Conclusion Yiqi Fuzheng Sijunzi Decoction and Yupingfeng Powder may regulate the immune function by affecting the immune organs and regulating the expression of cytokines IFN-γ and IL-4.
