目的 通过检测哮喘患者外周血单个核细胞 (PBMCs)腺苷受体mRNA表达 ,探讨腺苷受体在哮喘发病中的作用。方法 用Ficoll液分离PBMCs ,采用逆转录 多聚酶联反应法和图象分析半定量法检测PBMCs腺苷受体mRNA。结果 正常人及哮喘患者PBMCs均表达A1、A2a、A2b、A3 腺苷受体 (A1AR、A2aAR、A2bAR、A3AR)mRNA ,以A2bARmRNA所占百分比最高 ;哮喘患者PBMCsA1AR、A2bAR、A3ARmRNA表达较正常人明显增加。哮喘患者PBMCsA3ARmRNA表达与第一秒用力呼气容积实测值占预计值的百分率 (FEV1% )呈负相关。结论 哮喘患者PBMCs表达A1AR、A2bAR、A3ARmRNA增加 ,以A2bARmRNA所占百分比最高 ,A3ARmRNA表达与气道阻塞程度相关
Objective To investigate the role of adenosine receptor in the pathogenesis of asthma by detecting the expression of adenosine receptor mRNA in peripheral blood mononuclear cells (PBMCs) of asthmatic patients. Methods PBMCs were isolated by Ficoll solution, and adenosine receptor mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) and semi-quantitative image analysis. Results The expression of A1, A2a, A2b and A3AR mRNA in PBMCs of normal and asthmatic patients was the highest, and the percentage of A2bAR mRNA was highest. The expression of A1AR, A2bAR and A3AR mRNA in PBMCs of asthmatic patients was significantly higher than that of normal subjects increase. The expression of PBMCs A3AR mRNA in asthmatic patients was negatively correlated with the predicted FEV1% of the first second forced expiratory volume. Conclusions The expression of A1AR, A2bAR and A3AR mRNA in PBMCs of asthmatic patients is increased, and the percentage of A2bAR mRNA is the highest. The expression of A3AR mRNA is correlated with the degree of airway obstruction
