目的 :探索大剂量照射后造血生长因子受体基因的表达规律。方法 :应用逆转录PCR技术 ,从小鼠骨髓造血组织中扩增出造血生长因子受体的目的片段 ,PCR产物与克隆载体连接后转化感受态大肠杆菌 ,阳性菌落提取质粒后进行序列测定。结果 :4种造血生长因子受体片段均扩增成功 ,PCR产物经测序证实。结论 :粒细胞集落刺激因子受体 (G_CSFR)、促红细胞生成素受体 (EPOR)、促血小板生成素受体 (c_mpl)和干细胞因子受体 (c_kit)cDNA片段的克隆成功 ,为从整体水平上研究照射后造血生长因子受体的表达规律打下了基础。
Objective: To explore the expression of hematopoietic growth factor receptor gene after high dose irradiation. Methods: The target fragment of hematopoietic growth factor receptor was amplified from mouse bone marrow hematopoietic tissue by reverse transcription polymerase chain reaction (PCR). The PCR product was ligated with the cloning vector and transformed into competent E. coli. The positive colonies were extracted and sequenced. Results: All four hematopoietic growth factor receptor fragments were successfully amplified. PCR products were confirmed by sequencing. Conclusion: The cloning of G_CSFR, EPOR, c_mpl and c_kit cDNA fragments has been successfully cloned. On the study of irradiation hematopoietic growth factor receptor expression laid the foundation.
