目的建立隔山香药材中异莳萝脑乙二醇的TLC鉴别及HPLC含量测定方法。方法采用TLC法,以三氯甲烷-丙酮(5∶1)为展开剂,10%硫酸乙醇溶液为显色剂,对隔山香药材中的异莳萝脑乙二醇进行鉴别。采用HPLC法测定隔山香药材中异莳萝脑乙二醇的含量,色谱柱:Diamonsil C18色谱柱(5μm,250 mm×4.6 mm);流动相为甲醇-水(32∶68);流速为1.0 m L·min-1;检测波长为210 nm;柱温为25℃;进样量:10μL。结果在优化色谱条件下薄层斑点清晰,分离效果好,方法耐用性佳;异莳萝脑乙二醇在0.0468~0.468μg范围内线性关系良好(r=0.9999),平均回收率为101.4%,RSD为1.4%。结论本研究所建立的定性定量方法简便、准确、重现性好,可用于隔山香药材的质量控制。
OBJECTIVE To establish TLC identification and HPLC method for determination of ethylene glycol in Dasanxiang herb. Methods TLC method was used to identify the isodorubicin ethylene glycol in Acorus officinalis L. with chloroform-acetone (5: 1) as developing solvent and 10% sulfuric acid as the reagent. The content of ethylene glycol in Isochrysis galbanum was determined by HPLC. Diamonsil C18 column (5 μm, 250 mm × 4.6 mm) was used as the mobile phase. The mobile phase was methanol-water (32:68). The flow rate was 1.0 m L · min-1; detection wavelength was 210 nm; column temperature was 25 ℃; injection volume: 10μL. Results The results showed that the TLC spots were clear and the separation effect was good under the optimal chromatographic conditions. The method was good in durability. The linear relationship was good (r = 0.9999) in the range of 0.0468-0.468 μg with the average recovery of 101.4% RSD was 1.4%. Conclusion The qualitative and quantitative method established in this study is simple, accurate and reproducible. It can be used for the quality control of Geishan Herbs.
